- Expanding the Kinetic-Regulatory Model of Escherichia coli for High-Value Compound Production from Aromatic Amino AcidsPublication . Platon, Grigore; Universidade NOVA de Lisboa. Instituto de Tecnologia Química e Biológica António XavierAromatic amino acid-derived compounds have significant applications in nutrition and pharmaceutical industries. Dopamine, a tyrosine-derived neurotransmitter with critical roles in motor control and neurological function, serves primarily as a vasopressor for treating acute cardiovascular anomalies recently, with potential applications in treatment of Parkinson’s disease. Additionally, it serves as precursor for polydopamine, a versatile coating material with diverse industrial applications. This expanding potential drives demand for efficient and sustainable microbial production methods. This work investigates optimization strategies for dopamine biosynthesis in Escherichia coli using kinetic modeling.
- DESIGN AND CHARACTERIZATION OF MHETASE MUTANTS FOR IMPROVED ACTIVITY AND SOLUBILITY IN PET DEGRADATIONPublication . Ilunga, Constança Isabel Morgado; Universidade NOVA de Lisboa. Instituto de Tecnologia Química e Biológica António XavierPolyethylene terephthalate (PET) is a plastic polymer valued for its durability and chemical inertness. However, inefficient recycling and improper disposal pose major environmental challenges. The bacterium Ideonella sakaiensis can biodegrade PET with two enzymes: PETase, which cleaves PET into smaller intermediates like mono-(2-hydroxyethyl) terephthalate(MHET),and MHETase, which hydrolyzes MHET into the monomers terephthalic acid (TPA) and ethylene glycol (EG). While PETase has been widely studied and engineered, MHETase remains comparative ly underexplored. This work addresses this gap by engineering mutants to improve both its catalytical efficiency and solubility.
