S. aureus has developed resistance to virtually all known classes of antibiotics, therefore there is a need for the development of new therapies and further studies to better understand the Mode of Action (MOA) of new antibiotics. To contribute towards this goal, we aimed to construct a library of promoters and to study their response to the presence of a range of different antibiotics. To achieve this, S. aureus genomic DNA fragments were cloned into a plasmid vector, upstream of the gene encoding the fluorescent mNeonGreen protein. Strains containing the resulting transcriptional fusions were exposed to different antibiotics at varying concentrations.(...)"

However, despite the growing interest in studying this bacteria, there are only a few tools currently available for the genetic manipulation of S. aureus. In this study we developed genetic tools that could facilitate DNA insertion and gene repression in S. aureus: pKILL plasmid and CRISPRi.(...)"
