Resumo: "Organismsandenzymesarecurrentlyusedforcatalyzingtheproductionofvalue- added compounds,indiverseareaswitheconomicimpact.However,poorprotein expression, lowcatalyticlevels,lowconcentrationsofco-factorsorsubstrates,toxicity towardsthefinalproductandotherfactors,allcontributetothelowperformanceof microorganismsandenzymesintheseareas.Proteinengineeringisaneffectivestrategy forimprovingbiosyntheticpathwaysandovercomingtheseproblems,byre-designingan enzyme’scatalyticpropertiesinfavorofacertainreaction. Toaddressthisproblem,theProteinModelingLaboratoryofITQBNOVAdeveloped an automatedplatformforgenediscoveryandenzymeengineering,whichaimstoenhance or changetheenzymesthatcatalyzethelimitingstepsinpathwaysthatcanbeexploited fortheproductionofaddedvaluecompounds.Theobjectivesrangedfromincreasing an enzyme’sefficiencytoenablingittocatalyzeadifferenttransformationthantheone catalyzedbythewild-typesequence.(...)"
