Síntese e Caracterização de Sistemas Anfifílicos Multi-Responsivos

Nas últimas décadas, o desenvolvimento de novos sistemas para entrega controlada de fármacos tem sido intensamente investigado pela comunidade científica. Uma das estratégias consiste na utilização de agregados supramoleculares com a capacidade de encapsular e libertar as moléculas de interesse. Pretende-se desta forma melhorar a biodisponibilidade de fármacos, bem como a sua solubilidade e capacidade de penetração de membranas e, por outro, controlar a sua atuação in loco através da ação de estímulos externos ou endógenos (pH, luz, temperatura, etc). Este tipo de agregados pode ser construído tirando partido do efeito hidrofóbico e das interações não covalentes que se estabelecem entre compostos anfifílicos responsivos. A resposta combinada a estímulos de luz e pH apresenta vantagens do ponto de vista terapêutico, já que possibilita uma maior seletividade para células tumorais, cujo pH extracelular é mais acídico que nas células normais correspondentes. A família das antocianinas, moléculas abundantes na natureza, apresenta um sistema multiestado capaz de operar como um fotocomutador sensível ao pH (catião avílio - chalcona trans), o que o torna interessante para as aplicações acima descritas. No decorrer deste trabalho sintetizaram-se 2-hidroxichalconas de natureza anfifílica funcionalizadas com um grupo polar carregado positiva ou negativamente. Para isso foram usados os grupos tetrametiletilenodiamónio ou sulfonato, respetivamente, tendo-se conseguido obter estes compostos com bons rendimentos, na ordem dos 60-90 %. Todos os compostos foram caracterizados na forma monomérica por técnicas espetroscópicas, a vários valores de pH, tanto no equilíbrio térmico como no estado fotoestacionário. A capacidade de agregação foi investigada por tensiometria e ressonância magnética nuclear e, no caso do composto contendo um centro quiral, por dicroísmo circular. No caso dos compostos funcionalizados com o grupo substituinte aniónico, sintetizaram-se vários análogos com diferentes tamanhos de cadeia hidrofóbica, verificando-se um aumento da capacidade de agregação com o aumento do caráter hidrofóbico. Os compostos funcionalizados com o grupo substituinte catiónico incluem um tensioativo do tipo gemini e o respetivo análogo de cauda única, que serviu de composto modelo para o estudo das interações do primeiro. As interações recetor-substrato com um recetor macrocíclico solúvel em água (cucurbit[7]urilo) foram exploradas como estímulo adicional para controlar a agregação dos compostos sintetizados, bem como para modular as propriedades cinéticas e termodinâmicas dos sistemas multiestado.
Modulation of tumor microenvironment towards cancer therapy

Over the recent years, the role of the tumor microenvironment (TME) in tumor formation and progression has become increasingly recognized. Three-dimensional tumor models, such as spheroids, are powerful tools with great potential in preclinical research, as they recapitulate an array of TME features. While conventional therapies are effective in many cases, resistance phenomena and the occurrence of severe side effects are recurrent. Hence, it is of utmost importance the development of therapeutic approaches (e.g., combination therapies) that enable the reduction of the administered drug dosages, improving the therapeutic efficacy and decreasing the occurrence of side effects. In this thesis, colorectal cancer (CRC) 3D models were developed by using cells from CRC- derived cell lines alone or cocultured with fibroblasts, and their characterization was performed. Subsequently, functionalization of gold nanoparticles (AuNPs) with polyethylene glycol (PEG) was conducted, and the nanoconjugates obtained were characterized. Lastly, the dispersion of the nanoconjugates in spheroids was evaluated, as well as the effect that their irradiation caused on doxorubicin (Dox) diffusion. Characterization of monoculture models demonstrated the formation of a necrotic core. In coculture models, it was observed that the use of higher proportions of fibroblasts led to an increase in their dispersion in the 3D structure. Gold nanoparticles functionalization resulted in alterations in their surface chemistry, surface charge, and mean size. AuNPs dispersion in spheroids revealed the internalization of nanoconjugates through the formation of cell vacuoles. Preliminary results demonstrate a synergistic effect of AuNPs on Dox diffusion. Overall, the results obtained in this thesis unveil a potential application of these models in preclinical research, being preponderant in revolutionizing modern medicine. The great advantage of using AuNPs in combination therapies was also demonstrated.
The Use of Flavylium Salts as Dynamic Inhibitor Moieties for Human Cb5R

Cytochrome b5 reductase (Cb5R) is a flavoprotein that participates in the reduction of multiple biological redox partners. Co-localization of this protein with nitric oxide sources has been observed in neurons. In addition, the generation of superoxide anion radical by Cb5R has been observed. A search for specific inhibitors of Cb5R to understand the role of this protein in these new functions has been initiated. Previous studies have shown the ability of different flavonoids to inhibit Cb5R. Anthocyanins are a subgroup of flavonoids responsible for most red and blue colors found in flowers and fruits. Although usually represented by the flavylium cation form, these species are only stable at rather acidic pH values (pH ≤ 1). At higher pH values, the flavylium cation is involved in a dynamic reaction network comprising different neutral species with the potential ability to inhibit the activities of Cb5R. This study aims to provide insights into the molecular mechanism of interaction between flavonoids and Cb5R using flavylium salts as dynamic inhibitors. The outcome of this study might lead to the design of improved specific enzyme inhibitors in the future.
