Determination of the chemical composition of Eucalyptus spp. For cellulosic pulp production

The chemical composition of wood is important to assess the quality of this raw material for the industry of cellulosic pulp production. The purpose of this work was to determine the chemical composition of Eucalyptus spp. grown for cellulosic pulp production. Ten Eucalyptus spp. clones with six years of age, located in the municipality of Itamarandiba, Minas Gerais, Brazil, were used. Quantification was obtained for extractives, monosaccharides, uronic acids, acetates, lignin, ash and the phenolic composition of the extracts. In average, clones showed around 2.7% extractives, with a predominance of polar compounds soluble in ethanol and water; 27.7% lignin and 0.3% ash. Glucose was the main sugar detected (64.2%), followed by xylose (19.3%). The main components of the extractives were steroids, fatty acids and aromatic acids, followed by smaller amounts of substituted alkanoic acids, fatty alcohols, glycerol derivatives and triterpenes. The ethanol–water extracts presented total phenol contents ranging from 321.4 to 586.6 mg EAG/g of extract, tannins from 28.1 to 65.1 mg catechin/g of extract and flavonoids from 73.6 to 256.9 mg catechin/g of extract. Clones with a higher holocellulose amount and a lower lignin content showed a higher potential for cellulosic pulp production. These findings are important for the development of high-quality wood based on important traits for the pulp and paper sector.
Synthesis of glutathione as a central aspect of PAH toxicity in liver cells

Polycyclic Aromatic Hydrocarbons (PAH) are a class of organic pollutants normally found as mixtures with effects often hard to predict, which poses a major challenge for risk assessment. In this study, we address the effects of Phenanthrene (Phe), benzo[b]fluoranthene (B[b]F) and their mixtures (2 Phe:1 B[b]F; 1 Phe: 1 B[b]F; 1 Phe: 2 B[b]F) over glutathione (GSH) synthesis and function in HepG2 cells. We analyzed the effects on cellular viability, ROS production, glutathione (GSH) levels, protein-S-glutathionylation (PSSG), the activity of glutathione peroxidase (GPx), glutathione-S-transferases (GST) and glutathione reductase (GR). Transcript (mRNA) levels of glutathione synthesis enzymes - glutathione cysteine ligase catalytical (GCLC) and modifying (GCLM) sub-units and glutathione synthetase (GS) – and Nrf2 translocation to the nucleus were analyzed. Phe showed a higher cytotoxicity (IC50 = 130 µM after 24 h) than B[b]F related to a higher ROS production (up-to 50% for Phe). In agreement, GSH levels were significantly increased (up-to 3-fold) by B[b]F and were accompanied by an increase in the levels of PSSG, which is a mechanism that protect proteins from oxidative damage. The upregulation of GSH was the consequence of Nrf2 signaling activation and increased levels of GCLC, GCLM and GS mRNA observed after exposure to B[b]F, but not during exposure to Phe. Most interestingly, all mixtures showed higher cytotoxicity than individual compounds, but intriguingly it was the 1 Phe: 1B[b]F mixture showing the highest cytotoxicity and ROS production. GSH levels were not significantly upregulated not even in the mixture enriched in B[b]F. These results point to the role of GSH as a central modulator of PAH toxicity and demonstrate the idiosyncratic behavior of PAH mixtures even when considering only two compounds in varying ratios.
Investigations of olive oil industry by-products extracts with potential skin benefits in topical formulations

The by-products of olive oil industry are a major ecological issue due to their phenolic content, highly toxic organic load, and low pH. However, they can be recovered and reused, since their components have antioxidant, anti-inflammatory, and photoprotector properties. In this work, oil-in-water creams containing three different olive oil industry by-products extracts were produced without the use of organic solvents. First, the extracts were thoroughly characterized in vitro for cytotoxicity, inhibition of skin enzymes, and antioxidant and photoprotection capacities. Safety studies were then performed, including ocular and skin irritation tests, ecotoxicity evaluation, and in vivo Human Repeat Insult Patch Test. The results obtained in this initial characterization supported the incorporation of the extracts in the cream formulations. After preparation, the creams were characterized for their organoleptic, physicochemical, droplet size and rheological properties, and microbial contamination. The results showed that all formulations were semi-solid creams, with stable pH, compatible with the skin, without microbial contamination, and with the expected droplet size range. The rheological analysis showed shear-thinning behavior with yield stress, with the viscosity decreasing with increasing shear rate. The oscillatory results suggest that the creams have a strong network structure, being easily rubbed into the skin. Finally, compatibility, acceptability and antioxidant efficacy were evaluated in vivo, in human volunteers. No adverse reactions were observed after application of the formulations on skin and the cream with the highest concentrations of phenolic compounds showed the highest antioxidant efficiency. In conclusion, the results suggest that olive oil industry by-products extracts have valuable properties that favor their re-use in the cosmetic industry. The example presented here showed their successful incorporation into creams and their impact in these formulations’ appearance, pH, and rheological performance, as well as their in vivo compatibility with skin and antioxidant efficiency.
Green synthesis and photocatalytic dye degradation activity of CuO Nanoparticles

The degradation of dyes is a difficult task due to their persistent and stable nature; therefore, developing materials with desirable properties to degrade dyes is an important area of research. In the present study, we propose a simple, one-pot mechanochemical approach to synthesize CuO nanoparticles (NPs) using the leaf extract of Seriphidium oliverianum, as a reducing and stabilizing agent. The CuO NPs were characterized via X-ray diffraction (XRD), scanning electron microscopy (SEM), photoluminescence (PL) and Fourier-transform infrared spectroscopy (FTIR). The photocatalytic activity of CuO NPs was monitored using ultraviolet-visible (UV-Vis) spectroscopy. The CuO NPs exhibited high potential for the degradation of water-soluble industrial dyes. The degradation rates for methyl green (MG) and methyl orange (MO) were 65.231% ± 0.242 and 65.078% ± 0.392, respectively. Hybrid nanocomposites of Fe3O4/SiO2-EDTA

Fe3O4/SiO2-EDTA materials have been used for diverse applications and they have been modification by different methods, but a comparison is lacking between their physicochemical characteristics. This work aimed to compare the physicochemical characteristics of the Fe3O4/SiO2-EDTA nanocomposite obtained by four methods, whose modification of the Fe3O4 surface with SiO2-EDTA occurs in one or two steps, with not hazardous chemicals. In the one-step methods (M1 and M2), Fe3O4 particles were coated and functionalized in consecutive steps (by sonication in M1 and mechanical stirring in M2). In the two-step methods (M3 and M4), the Fe3O4 particles were primarily coated with silica and then functionalized with TMS-EDTA (by sonication in M3 and mechanical stirring in M4). The Fe3O4/SiO2-EDTA nanocomposites were characterized by XRD, TEM, EDX, BET, SQUID magnetometry, FTIR, XPS, and zeta potential. The TEM highlighted that the nanocomposites from methods M1 and M2 there is the simultaneous presence of magnetite particles coated and uncoated with silica, while those from M3 and M4 were totally coated, besides the nanocomposites from M2 and M3 showed to be more aggregated than those from the other methods. The presence of EDTA on the surface of the nanocomposites was evidenced by the carboxyl groups in the FTIR spectra and by the nitrogen in the XPS spectra. The nanocomposites from M3 and M4 presented a higher content of nitrogen than those from M1 and M2, and those from M4 also presented a higher content of carbon than those from M3. Overall, the Fe3O4/SiO2-EDTA nanocomposites from two-step methods showed better SiO2 coating and EDTA functionalization than those from one-step methods, and those from M4 presented the best physicochemical characteristics, being this method recommended for their modification for future applications.
