As doenças autoimunes são cada vez mais prevalentes. O Lúpus Eritematoso Sistémico (LES) é exemplo disso, apresentando maior prevalência no sexo feminino. Estas patologias caracterizam-se por uma atividade aberrante do sistema imunitário e uma inflamação sistémica, também denominada metainflamação. Esta última está também presente noutras patologias como a obesidade ou a diabetes. Este estudo consiste numa análise retrospetiva secundária de dados de 42 indivíduos do sexo feminino com LES pertencentes ao ensaio clínico METAINFLAMACION-CM (ref.Y2020/6600). Analisaram-se dados antropométricos, bioquímicos gerais e inflamatórios, e metagenómicos com o objetivo de compreender o papel da microbiota na perda de peso. Para isso a amostra foi dividida em dois grupos de acordo com a quantidade de peso perdido (< ou ≥ 3,5Kg) e realizaram se análises de diversidade bacteriana da microbiota intestinal, comparação entre grupos, associação, correlação e hierarquia taxonómica. Surgiram 9 géneros bacterianos candidatos a preditor para perda de peso nesta população, servindo este trabalho como uma prova de conceito, ao demonstrar a influência da microbiota na gestão de peso e de inflamação nos indivíduos com estas patologias. O presente estudo vem assim demonstrar a importância da medicina de precisão para o desenvolvimento de estratégias de gestão destas patologias crónicas caracterizadas por metainflamação.
The prevalence of autoimmune diseases is increasing, and Systemic Lupus Erythematosus (LES) is one example. It is more prevalent in women. These pathologies are characterized by aberrant immune system activity and systemic inflammation, also called metainflammation. The latter is also present in other pathologies such as obesity or diabetes. This study consists of a retrospective secondary analysis of data from 42 female individuals with LES belonging to the METAINFLAMACION-CM clinical trial (ref. Y2020/6600). Anthropometric, general and inflammatory biochemical, and metagenomic data were analysed to understand the role of the microbiota in weight loss. For this purpose, the sample was divided into two groups according to the amount of weight lost (< or ≥ 3,5 kg) and analyses of bacterial diversity of the intestinal microbiota, comparison between groups, association, correlation and taxonomic hierarchy were performed. Nine bacterial genera emerged as potential predictors of weight loss in this population. Therefore, this work serves as a proof of concept, demonstrating the influence of the microbiota on weight management and inflammation in individuals with i these pathologies. This study thus demonstrates the importance of precision medicine for the development of management strategies for these chronic pathologies characterized by metainflammation.
