Study of human gut microbiome dynamics in general population As part of the Citizen Science project Microbioma Comunidade Portugal

Resumo(s)

The human gut microbiome is highly diverse and plays a crucial role in health, influenced by factors such as diet, environment, and genetics. Although its composition has been extensively studied in several countries, the Portuguese microbiome remains poorly characterized. Moreover, only recently have a few longitudinal studies begun to emerge. This gap is addressed in this thesis through Microbioma Comunidade Portugal, a citizen science project involving 32 families, mainly from Oeiras municipality. Participants collected three fecal samples over time, and the family-based design allowed the inclusion of individuals across different ages and contributed to sustained motivation. Results revealed that these Portuguese cohort’s gut microbiota was dominated by Bacillota and Bacteroidota, consistent with other studies, and remained stable over time at the phylum level. The Bacillota/Bacteroidota ratio was consistent across time, age, and body mass index. Interestingly, alpha diversity increased overtime, possibly reflecting seasonal and/or behavior changes of the participants. Surprisingly, alpha diversity was higher in the elderly. Similar to other studies, intraindividual diversity was lower than interindividual diversity. Fecal metabolomic profiling did not show notable changes over time, and no correlations were found between key short-chain fatty acid-producing genera and the concentrations of these metabolites. Participants actively contributed feedback and engaged in laboratory visits and events, enhancing scientific literacy and trust in science. This study constitutes the first longitudinal analysis of the Portuguese gut microbiome, with an adherence rate of 94%, highlighting the valuable role of citizen science in microbiome research. Future studies involving larger and more geographically diverse cohorts will be essential to clarify the observed alpha diversity variations and further explore the functional dynamics of the gut microbiome
O microbioma intestinal humano é altamente diverso e desempenha um papel crucial na saúde, sendo influenciada por fatores como dieta, ambiente e genética. Embora a sua composição tenha sido amplamente estudada em vários países, o microbioma português permanece pouco caracterizado. Além disso, apenas recentemente alguns estudos longitudinais começaram a surgir. Estas lacunas são colmatadas nesta tese através do Microbioma Comunidade Portugal, um projeto de ciência cidadã que envolveu 32 famílias, maioritariamente do concelho de Oeiras. Os participantes recolheram três amostras fecais ao longo do tempo e o desenho familiar permitiu incluir indivíduos de diferentes idades, contribuindo para uma elevada adesão e motivação contínua. Os resultados mostraram que a microbiota intestinal desta coorte portuguesa foi dominada pelos filos Bacillota e Bacteroidota, em concordância com outros estudos, mantendo-se estável ao longo do tempo ao nível do filo. A razão Bacillota/Bacteroidota manteve-se consistente ao longo do tempo, idade e índice de massa corporal. Interessantemente, a diversidade alfa aumentou ao longo do tempo, possivelmente refletindo alterações sazonais e/ou comportamentais dos participantes. Surpreendentemente, a diversidade alfa foi maior nos idosos. Assim como em outros estudos, a diversidade intraindividual foi menor que a diversidade interindividual. A análise metabolómica das fezes não revelou alterações significativas ao longo do tempo, nem correlações entre os géneros produtores de ácidos gordos de cadeia curta e as concentrações destes metabolitos. Os participantes contribuíram com feedback e envolveram-se em visitas laboratoriais e eventos, promovendo a literacia científica e a confiança na ciência. Este estudo representa a primeira análise longitudinal do microbioma intestinal português, com uma taxa de adesão de 94%, destacando o valor da ciência cidadã na investigação do microbioma. Estudos futuros com coortes maiores e geograficamente mais diversas serão essenciais para clarificar as variações observadas na diversidade alfa e aprofundar a compreensão da dinâmica do microbioma intestinal.
